Technology Purtein has developed a novel protein purification system. It is a new TAG based on a specific protein with unique binding properties and affinity for commonly used Triazine dyes. Our proprietary technology is optimized for fusion protein recovery and purity in an inexpensive, rapid two-step column chromatography system. The initial phase uses a gravity flow column consisting of a Triazine dye immobilized on agarose beads. The second phase follows with rapid flow hydrophobic interaction column chromatography. The Purtein fusion protein is desalted, neutralized, and eluted in water during this two-step process. Advantages of the Purtein system include: - It can be produced in E.coli, Chinese hamster ovary cells, and bacculovirus insect cell systems;
- The procedures and conditions for the Purtein purification system are the same, regardless of which fusion protein production system used;
- The tracking and quantification of the Purtein tag, thus the fusion protein, can be easily assayed by a sensitive antibody capture ELISA;
- The binding and elution profile of Purtein’s tag is not affected by anti-foaming agents; and the system is highly reproducible and requires little or no troubleshooting.
Our technology covers two main applications: 1) Purification of the superfamily of proteins that contain a particular structural topology, a motif found in a wide variety of naturally occurring human proteins. These include growth factors and hormones of therapeutic and high commercial value. 2) The purification of almost any soluble recombinant fusion protein composed of a TAG formed from a fragment of a protein from the above family and the protein of interest. A recombinant fusion protein consisting of a protein of interest fused to the TAG protein is constructed and purified using the same method used for naturally occurring proteins from the superfamily. | |
